Alkylating Agents

The analysis of alkylating agents is required in fields such as environmental studies, chemical weapons, toxicology, oncology, and pharmaceutical quality control.

An alkylating agent is a compound that substitutes an alkyl group, CnH2n+1, for an active hydrogen atom in an organic compound. All alkylating agents form strong electrophiles through the formation of carbonium ion intermediates. This results in the formation of covalent linkages by alkylation of various nucleophile moieties. Their chemotherapeutic and cytotoxic effects are directly related to the alkylation of DNA. The formation of one covalent bond with nucleophiles can result in mutagenesis or teratogenesis but the formation of two of these bonds through cross-linking can produce cytotoxicity.

To cite a few examples, most acrylates (salts and esters of acrylic acid incorporating the acrylate ion having an anionic formula of CH2=CHCOO-) and mesylates (a salt or ester of methanesulfonic acid having an anionic formula of CH3SO3-) are excellent alkylating agents and are often used as synthetic building blocks.

As a result of this interaction, it is extremely important to have proof that all precursor alkylating agents are either completely consumed or removed prior to producing the final Active Pharmaceutical Ingredient or Drug Product.

The determination of residual alkylating reagents in pharmaceuticals is currently attracting the interest of regulatory authorities, both in Europe and in the United States. Although European guidelines have been recently established limiting concentrations to values below 1.5 ppm (1.5 ug/g), information in the low ppb (ng/g) range is highly desirable.
